Poly[bis[μ3-2-(1H-tetrazol-1-yl)acetato]cadmium(II)]
In the title compound, [Cd(C3H3N4O2)2]n, the CdII ion, located on a twofold rotation axis, is six-coordinated by two N atoms [Cd—N = 2.368 (2) Å] and four O atoms [Cd—O = 2.300 (1) and 2.260 (1) Å] from six 2-(1H-tetrazol-1-yl)acetate (L) ligands in a distorted octahedral geometry. The metal centres are connected via the tridentate L ligands into a three-dimensional polymeric structure
Orexin Regulates Bone Remodeling via a Dominant Positive Central Action and a Subordinate Negative Peripheral Action
SummaryOrexin neuropeptides promote arousal, appetite, reward, and energy expenditure. However, whether orexin affects bone mass accrual is unknown. Here, we show that orexin functions centrally through orexin receptor 2 (OX2R) in the brain to enhance bone formation. OX2R null mice exhibit low bone mass owing to elevated circulating leptin, whereas central administration of an OX2R-selective agonist augments bone mass. Conversely, orexin also functions peripherally through orexin receptor 1 (OX1R) in the bone to suppress bone formation. OX1R null mice exhibit high bone mass owing to a differentiation shift from marrow adipocyte to osteoblast that results from higher osseous ghrelin expression. The central action is dominant because bone mass is reduced in orexin null and OX1R2R double null mice but enhanced in orexin-overexpressing transgenic mice. These findings reveal orexin as a critical rheostat of skeletal homeostasis that exerts a yin-yang dual regulation and highlight orexin as a therapeutic target for osteoporosis

Quantum size effects on the perpendicular upper critical field in ultra-thin lead films
We report the thickness-dependent (in terms of atomic layers) oscillation
behavior of the perpendicular upper critical field in the
ultra-thin lead films at the reduced temperature (). Distinct
oscillations of the normal-state resistivity as a function of film thickness
have also been observed. Compared with the oscillation, the
shows a considerable large oscillation amplitude and a phase shift. The
oscillatory mean free path caused by quantum size effect plays a role in
oscillation.Comment: 4 pages, 4 figure
Periodontal Disease and Risk of Bladder Cancer: A Meta-Analysis of 298476 Participants
Objective: It has been reported that the periodontal disease is linked to a number of malignant tumors such as lung cancer and pancreatic cancer. In this study, we aimed to investigate the association of periodontal disease with risk of bladder cancer by a meta-analysis.Methods: PubMed, Scopus, ScienceDirect, and Chinese National Knowledge Infrastructure (CNKI) were searched for eligible publications up to December 15, 2017. Cohort and nested case-control studies on the association between periodontal disease and risk of bladder cancer were included. After study selection and data extraction, pooled hazard ratios (HRs) and their 95% confidence intervals (95%CIs) were calculated using a fixed-effect inverse-variance model. All analyses were performed using the RevMan 5.3 software. Results: Finally, five cohort studies were identified and included in this meta-analysis, involving 1,104 bladder cancer cases of 298,476 participants. Summary estimates based on adjusted data showed that periodontal disease was not significantly associated with the risk of bladder cancer (HR = 1.09, 95% CI = 0.95–1.25, I2 = 0%). A similar result was also observed after cumulative, subgroup and sensitivity analyses. Conclusions: Current evidence from cohort studies suggests that patients with periodontal disease may not be at an increased risk of developing bladder cancer
Influence of Surgical Technique on Mastectomy and Reexcision Rates in Breast-Conserving Therapy for Cancer
Introduction. Breast conserving surgery (BCS) requires tumor excision with negative margins. Reexcision rates of 30–50% are reported. Ultrasound localization, intraoperative margin pathology, and specimen mammography have reduced reexcisions, but require new equipment. Cavity shave margin (CSM) is a technique, utilizing existing equipment, that potentially reduces reexcision. This study evaluates CSM reexcision impact. Methods. 522 cancers treated with BCS were reviewed. Patients underwent standard partial mastectomy (SPM) or CSM. Data collected included demographics, pathology, and treatments. Results. 455 SPMs were compared to 67 CSMs. Analysis revealed no differences in pathology, intraductal component, or neoadjuvant chemotherapy. Overall reexcision rate = 43%. Most reexcisions were performed for DCIS at margin. SPMs underwent 213 reexcisions (46.8%), versus 16/67 (23.9%) CSMs (P = 0.0003). Total mastectomy as definitive procedure was performed after more SPMs (P = 0.009). Multivariate analysis revealed CSM, % DCIS, tumor size, and race to influence reexcisions. Conclusions. CSM is a technique that reduces reexcisions and mastectomy rates
NQO1-dependent, tumor-selective radiosensitization of non-small cell lung cancers
Purpose:
Development of tumor-specific therapies for the treatment of recalcitrant non-small cell lung cancers (NSCLCs) are urgently needed. Here, we investigated the ability of ß-lapachone (ß-lap, ARQ761 in clinical form) to selectively potentiate the effects of ionizing radiation (IR, 1–3 Gy) in NSCLCs that over-express NAD(P)H:Quinone Oxidoreductase 1 (NQO1).
Experimental Design:
The mechanism of lethality of low dose IR in combination with sublethal doses of ß-lap were evaluated in NSCLC lines in vitro and validated in subcutaneous and orthotopic xenograph models in vivo. Pharmacokinetics and pharmacodynamics (PK/PD) studies comparing single versus co-treatments were performed to validate therapeutic efficacy and mechanism of action.
Results:
ß-Lap administration after IR treatment hyperactivated PARP, greatly lowered NAD+/ATP levels, and increased DSB lesions over time in vitro. Radiosensitization of orthotopic, as well as subcutaneous, NSCLCs occurred with high apparent cures (>70%), even though 1/8 ß-lap doses reach subcutaneous versus orthotopic tumors. No methemoglobinemia or long-term toxicities were noted in any normal tissues, including mouse liver that expresses the highest level of NQO1 (~12 Units) of any normal tissue. PK/PD responses confirm that IR + ß-lap treatments hyperactivate PARP activity, greatly lower NAD+/ATP levels and dramatically inhibit DSB repair in exposed NQO1+ cancer tissue, while low NQO1 levels and high levels of Catalase in associated normal tissue were protective.
Conclusion:
Our data suggest that combination of sublethal doses of ß-lap and IR is a viable approach to selectively treat NQO1-overexpressing NSCLC and warrant a clinical trial using low-dose IR + ß-lapachone against patients with NQO1+ NSCLCs
Phytochromes are Involved in Elongation of Seminal Roots and Accumulation of Dry substances in Rice Seedlings
Abstract: Phytochromes have been reported to play important roles in seedling de-etiolation and flowering in rice. To identify the roles of phytochromes in regulating root growth and accumulation of dry substances, the lengths of seminal roots and dry weights of seedlings were measured in wild type as well as phytochrome A (phyA) and phytochrome B (phyB) mutants grown under different conditions. When the whole seedlings were exposed to white light, the elongation of the seminal roots was significantly photoinhibited in the wild type, whereas this inhibitory effect was clearly reduced in the phyA and phyB mutants. When the roots of the seedlings were blocked from white light, the phyA and phyB mutants exhibited significantly longer seminal roots than the wild type. These results suggest that both the root-localized and shoot-localized phyA and phyB are involved in the photoinhibition of seminal root elongation in rice seedlings. By measuring the dry weights of roots and shoots, it is revealed that phyB positively regulates the accumulation of dry substances in shoots. Moreover, we reveal that phyA plays the reverse effects on the accumulation of dry substances in roots and shoots in rice seedlings
